Summary:A treponema antigen is prepd. from a strain of Treponeuca Pallidum registered under No 13156 in the collection of the Cautacuzino Triotitute in Bucherest. This is injected in the testicles of rabbits. The infected tissues are collected after ten days and are suspended in a 100 i.u-s heparin/ml containing, phosphate-saline buffer. The suspn. is allowed to stand for 45 mins at 37 deg.C. The supernatant is decanted, centrifuged and filtered through a medium porosity fritted glass filter. The filtrate is treated by a mixt. comprising a methyl-glucamine salt of N, N'-di:acetyl -3-5-di:amino-2,4,6 -tri:iodobenzoic-acid and poly-dextran of 4,000,000 Mol.Wt.; followed by centrifuging. Formaldehyde is added to the supernatant and allowed to stand for 7 days at 4 deg.C prior to centrifuging. The sediment is rinsed with phosphate saline buffer. Final concn. of 2 x 10 power 7 germs/ml phosphate-buffer is set by centrifuging. 0.75% glycocol, 0.5% sodium-glutenate and 0.02% Thimerosal is added. The prepn. is proportioned in vials, and hyophilised. Frozen at -58 deg.C and dried for 48 hours up to 30 deg.C max. Closure of vial occurs in Ar atmos.
