Summary: | The present disclosure discloses a mouse T-cell-dependent antibody response (TDAR) assay kit, including a kit packaging, an antigen keyhole limpet hemocyanin (KLH), an antigen-coated enzyme-linked immunosorbent assay (ELISA) plate, a 5 biotin-labeled goat anti-mouse enzyme-labeled secondary antibody diluent, a purified anti-mouse KLH polyclonal IgG antibody, a sample diluent, a washing buffer, a 3,3',5,5'-tetramethylbenzidine (TMB) chromogenic solution, and a stop solution. An anti-mouse KLH polyclonal IgG antibody is purified through a combination of salt precipitation and affinity chromatography. A coating concentration of the KLH antigen 10 and a dilution ratio of the goat anti-mouse enzyme-labeled secondary antibody diluent are controlled, the purified anti-mouse KLH polyclonal IgG antibody is prepared, and KLH immune conditions and an effective dilution ratio of a test sample are designed to make the overall detection system more adaptable to the effective analysis of a TDAR experimental result through a standard concentration and effectively realize the 15 transformation of TDAR from qualitative assay to quantitative assay, with high detection sensitivity, strong specificity, convenient operation, and high practicability. 1 |
