Summary: | The present invention discloses a colorimetric and fluorometric detection system based on the fluorometric or calorimetric loop-mediated isothermal reverse transcription (RT-LAMP) DNA amplification technique to precisely detect the SARS-CoV-2 virus in situ, preferably on saliva samples with or without RNA extraction, which uses recombinant enzymes Bst DNA polymerase and reverse transcriptase, and hybridization oligonucleotides corresponding to the N1 gene of SARS-CoV-2 selected in sequence and quantity. Also, it discloses its development and aims to provide its proof of concept.
La presente invención describe un sistema de detección colorimétrica y fluorom trica basado en la técnica de amplificación de DNA por transcripción reversa isotérmica mediada por loops (RT-LAMP) fluorométrico o calorimétrico, para detectar específicamente al virus SARS-CoV-2 in sitio, preferentemente para realizarse en muestras de saliva con o sin extracción de RNA, el cual utiliza enzimas recombinantes Bst DNA polimerasa y reverso transcriptasa, así como oligonucleótidos de hibridación correspondientes al gen N1 de SARS-CoV-2 seleccionados en secuencia y en cantidad; así mismo se describe el desarrollo detallado de la misma y se proporciona la prueba de concepto. |
