Summary:The current invention involves a series of fully recombinant multimerized forms of immunoglobulin Fc which thereby present polyvalent immunoglobulin Fc to immune cell receptors. The fusion proteins exist as both homodimeric and highly ordered multimeric fractions, termed stradomers. In comparison to the homodimeric fraction, purified multimeric stradomers have higher affinity and avidity for Fc Rs with slower dissociation and are useful in the treatment and prevention of disease. The current invention demonstrates that directly linking IgG1 Fc regions to multimerization domains leads to enhanced multimerization and biological activity. La presente invención implica una serie de formas multimerizadas completamente recombinantes de Fc de inmunoglobulina que así presenta el Fc de inmunoglobulina polivalente a los receptores de las células inmune. Las proteínas de fusión existen tanto homodiméricas y fracciones multiméricas altamente ordenadas, denominadas estradómeros. En comparación con la fracción homodimérica, los estradómeros multiméricos purificados tienen mayor afinidad y avidez por los Fc?R con disociación más lenta y son útiles en el tratamiento y prevención de enfermedades. La presente invención demuestra que enlazar directamente las regiones Fc de IgG1 a dominios de multimerización conduce a multimerización y actividad biológica mejoradas.
