Summary:L-Tryptophan is prepd. from L- or DL-serine, indole, and ammonium salts enzymatically in nonaqueous solvent systems. Thus, a mixt. of L-serine 9.27, (NH4)2 SO4 3, pyridoxal phosphate 0.01 and H2O 82.5g is adjusted to pH8.5, mixed with E. coli MT10242 cream cake 3 and acetyltributyl citrate (contg, indole) 68.9g. The reaction mixt. is incubated at 35 C for 2hr to give 96.5% Ltryptophan based on the indole used. Nonaqueous onganic solvent contains toluene, methyl iso- ketone, diisoketone, diiso butyl ketone, ethyl citrate, and benzene.
