A METHOD FOR STABILIZING A TUMOR NECTROSIS FACTOR

A tumor necrosis factor(TNF) compn. was stabilized in aq. soln. or powder form by the addn. of at least one stabilizer(I) selected from albumin, gelation, protamines and protamine salts. Thus, a soln. of rabbit TNF(sp. activity; 5.0 x105 u/mg) in 0.15M NaCl was dild. with 0.1M phosphate buffer(pH 7....

Summary:A tumor necrosis factor(TNF) compn. was stabilized in aq. soln. or powder form by the addn. of at least one stabilizer(I) selected from albumin, gelation, protamines and protamine salts. Thus, a soln. of rabbit TNF(sp. activity; 5.0 x105 u/mg) in 0.15M NaCl was dild. with 0.1M phosphate buffer(pH 7.0) to form a TMF soln. with an activity of 1200 u/ml. HSA, BSA, HGG, and PS were sep. added to portion of TNF soln. to form 2 different solns(samples) having a concn. of 0.1 mg/ml or 1.0 mg/ml. The residual activity of the sample with a (I) was 87 - 100% after treatments while that of the control TNF sample without a (I) was 4 - 52%. (I) stabilizes TNF against loss of activity during storage, freeze-drying and freeze-thaw treatment. The stabilized soln. may be freeze dried to obtain compns. on powder form.
