Summary:The present invention provides a manufacturing method of a graphene composite structure, and a graphene composite structure manufactured thereby, which gives hydrophilicity to a graphene layer on which a biological sample is placed while reducing spontaneous light in an observation portion to enable high magnification and high clarity analysis of the biological sample. Disclosed in the present invention are a manufacturing method of a graphene composite structure, and a graphene composite structure manufactured thereby, wherein the manufacturing method comprises: a support forming step of forming a support having an observation portion corresponding to an observation region and a frame portion disposed on a border of the observation portion; a graphene layer forming step of forming a graphene layer on an upper surface of the observation portion; and a hydrophilic protective layer forming step of transferring an organic component to an upper surface of the graphene layer by allowing an organic elastic member to be in physical contact with an upper surface of the graphene layer, and applying energy to the graphene layer on which the organic component is transferred to form a hydrophilic protective layer on the upper surface of the graphene layer. 본 발명은 생물학적 샘플이 올려지는 그래핀층의 친수성을 부여하는 동시에 관찰부에서의 자발광을 감소하여 생물학적 샘플의 고배율 및 고선명도 분석이 가능하도록 하는 그래핀 복합 구조체 제조방법 및 이로부터 제조되는 그래핀 복합 구조체를 제공함에 있다. 이를 위한 본 발명은 관찰 영역에 해당하는 관찰부와 상기 관찰부의 테두리에 배치되는 프레임부를 가지는 지지체를 형성하는 지지체 형성단계; 상기 관찰부의 상면에 그래핀층을 형성하는 그래핀층 형성단계; 및 상기 그래핀층의 상면에 유기 탄성부재를 물리적으로 접촉시켜 상기 그래핀층의 상면에 유기성분을 전사하고, 상기 유기성분이 전사된 상기 그래핀층에 에너지를 인가하여 상기 그래핀층의 상면에 친수화 보호층을 형성하는 친수화 보호층 형성단계;를 포함하는 그래핀 복합 구조체 제조방법와 이로부터 제조된 그래핀 복합 구조체를 개시한다.
