Summary:In the present invention, 1-DNJ core biosynthesis gene group was isolated from Bacillus atrophaeus MBLB1156 (KCCM12922P) isolated from fermented foods and characteristics thereof were studied through the heterozygous expression of each gene. First, GabT1, a first biosynthetic enzyme, was found to prefer fructose-6-phosphate as an amino group acceptor and L-glutamic acid as an amino group donor as an aminotransferase in the presence of the coenzyme PLP. Yktc1, a second biosynthetic enzyme, was found to catalyze the dephosphorylation reaction with a zinc ion-dependent phosphatase, and the optimal activity condition was 65 ℃ and pH 5.0. In addition, Yktc1 was found to have a wide range of substrate specificities. GutB1, the last biosynthetic enzyme, has a high substrate specificity for 2-amino-2-deoxy-D-mannitol, and the optimum activity condition was found to be 30 ℃ and pH 9.5. The characteristic analysis of biosynthetic enzymes performed in the present invention can be used as basic data for identifying and understanding the biosynthetic pathway of 1-DNJ. 본 발명은 발효식품에서 분리된바실러스 아트로페어스 MBLB1156 (Bacillus atrophaeus MBLB1156)(KCCM12922P)으로부터 1-DNJ 핵심 생합성 유전자 군을 분리하여 각 유전자들의 이형발현을 통해 특성을 연구하였다. 먼저 첫 번째 생합성 효소인 GabT1은 조효소인 PLP 존재하에 아미노기전이효소로 아미노기 공여체로 L-글루탐산을 아미노기 수용체로 fructose-6-phosphate를 선호하는 것으로 확인되었다. 두 번째 생합성 효소 Yktc1은 zinc이온 의존성 포스파타아제로 탈인산화반응을 촉매하는 것으로 확인이 되었고 최적활성 조건은 65℃ pH 5.0 였다. 또한 Yktc1은 넓은 범위의 기질 특이성을 갖는 것으로 확인되었다. 마지막 생합성 효소 GutB1은 2-amino-2-deoxy-D-mannitol에 대해 높은 기질 특이성을 가지고 최적활성 조건은 30℃ pH 9.5로 확인되었다.
