Summary:The present invention relates to a DNA polymerase extracted and purified from Geobacillus lituanicus with properties similar to Bst DNA polymerase, in which a protein sequence of DNA polymerase I extracted from Geobacillus lituanicus was compared and inferred with Bst DNA polymerase extracted from Bacillus stearothermophilus, so as to biosynthesize 1,749 bases corresponding to C-terminus 583 amino acids related to the Bst DNA polymerase in DNA polymerase I extracted from the Geobacillus lituanicus, and clone the biosynthetic product into a bacterial expression vector. According to the present invention, a new resource is secured by extraction of DNA polymerase from a new Bacillus species to have similar efficacy to Bst DNA polymerase extracted from Bacillus stearothermophilus, instead of Bst DNA polymerase, and thus has the effect of providing a DNA polymerase which can be advantageously used when performing isothermal PCR. 본 발명은 지오바실러스 리투아니커스(Geobacillus lituanicus)로부터 추출한 DNA 폴리머라제 I을 바실러스 스테아로써모필루스(Bacillus stearothermophilus)에서 추출한 Bst DNA 폴리머라제와 단백질 서열을 비교 유추하여, 상기 지오바실러스 리투아니커스(Geobacillus lituanicus)로부터 추출한 DNA 폴리머라제 I에서 상기 Bst DNA 폴리머라제와 관계가 있는 C-terminus 583개의 아미노산에 해당하는 1,749개의 염기를 생합성하고, 상기 생합성한 결과물을 박테리아 발현 벡터(expression vector)에 클로닝한 것을 특징으로 하는, Bst DNA 폴리머라제와 유사 성질을 가진 지오바실러스 리투아니커스에서 추출 및 정제된 DNA 폴리머라제에 관한 것이다. 본 발명에 따르면, 바실러스 스테아로써모필루스에서 추출한 Bst DNA 폴리머라제와 유사한 효능을 가지도록 새로운 바실러스(Bacillus) 종에서 DNA 폴리머라제의 추출에 의해 새로운 자원의 확보를 통해서, Bst DNA 폴리머라제를 대신해서 등온 PCR을 실시할 때 유용하게 사용할 수 있는 DNA 폴리머라제를 제공하는 효과를 가진다.
