Summary:The present invention relates to a method for preparing a conjugate of S. pneumoniae serotype 23F with a carrier protein, including the steps of: (i) mixing capsular polysaccharides of S. pneumoniae serotype 23F with a sodium acetate (NaOAc) buffer to obtain a mixture; (ii) reacting the mixture with an oxidizing agent to produce activated serotype 23F polysaccharides; (iii) filtering the activated serotype 23F polysaccharides with sodium acetate to select activated serotype 23F polysaccharides having a molecular weight of at least 300 kDa; (iv) mixing the activated serotype 23F polysaccharides with a carrier protein; and (v) adding a reducing agent to the mixture of the activated serotype 23F polysaccharides with the carrier protein to perform conjugation and to form a conjugate of the activated serotype 23F polysaccharides with the carrier protein. According to the present invention, it is possible to maintain the size of capsular polysaccharides of S. pneumoniae serotype 23F constantly, and to provide a conjugate having excellent immunogenicity advantageously. 본 발명은 (i) 정제된 스트랩토코커스 뉴모니애(S. pneumoniae) 혈청형 23F의 협막 다당류를 아세트산 나트륨(NaOAc) 완충액에 혼합하여 혼합물을 얻는 단계, (ii) 상기 혼합물과 산화제를 반응시켜 활성화된 혈청형 23F 다당류를 생성시키는 단계, (iii) 활성화된 혈청형 23F 다당류를 아세트산 나트륨으로 여과시켜 적어도 300kDa 을 갖는 활성화된 혈청형 23F 다당류를 선택하는 단계, (iv) 활성화된 혈청형 23F 다당류를 운반체 단백질과 혼합하는 단계, 및 (v) 활성화된 혈청형 23F 다당류 및 운반체 단백질의 혼합물에 환원제를 첨가하여 컨쥬게이션시켜 활성화된 혈청형 23F 다당류 및 운반체 단백질의 접합체를 형성하는 단계를 포함하는, 스트랩토코커스 뉴모니애(S. pneumoniae) 혈청형 23F와 운반체 단백질의 접합체를 제조하는 방법을 제공하고자 한다. 본 발명은 스트랩토코커스 뉴모니애(S. pneumoniae) 혈청형 23F의 협막 다당류의 크기를 일정하게 유지할 수 있고, 우수한 면역원성을 갖는 접합체를 제공하는데 유리할 수 있다.
