Summary:The present invention relates to an optical based graphene biosensor using aptamers, a preparation method thereof, and a method for detecting a target material using the same. More specifically, according to the present invention, when a fluorescent aptamer forms a double helix structure with a complementary nucleic acid sequence thereof, and is fixated to a graphene substrate by a fixing material so that is exposed to the target material, the fluorescent material marked on the aptamer is reacted with the surface of graphene to lose the fluorescence while the fluorescent aptamer forms a single helix structure composite with the target material by the stronger attraction force than the complementary nucleic acid sequence thereof, thereby the target material can be quantitatively detected.
