PURPOSE: A serum-free and antibiotics-free culture medium composition and a method for preparing the same are provided to obtain the culture medium for mesenchymal stem cells and to be used in a cosmetic composition. CONSTITUTION: A cosmetic composition contains a culture medium for mesenchymal stem cells derived from bone marrow or umbilical cord blood of a mammal, which is treated with beta-defensin 3 recombinant protein. The culture medium is a serum-free medium. The beta-defensin 3 recombinant protein has an amino acid sequence of sequence number 1. A method for preparing the culture medium comprises: a step of culturing mesenchymal stem cells derived from isolated bone marrow or umbilical cord blood in a serum-containing medium; and a step of culturing the mesenchymal stem cells in a serum-free medium containing the beta-defensin 3 recombinant protein.
