Summary: | PURPOSE: A method for preparing insulin-secreting cells through reprogramming from labium minor-derived adult cells is provided to easily and rapidly obtain massive adult cells from labium minor. CONSTITUTION: A method for inducing the differentiation of pluripotent adult stem cells which are derived from labium minor to insulin-secreting cells comprises: a step of culturing the adult stem cells in the media containing low glucose DMEM, 10% of FBS and 1% L-glutamine and 1% of penicillin-streptomycin; a step of culturing cultured adult stem cell in ITS media containing DMEM/F12(1:1), 5μg/ml of insulin, 50ug/ml of transferrine, 30nM of selenium and 1mM of glutamine; and a step of culturing the cultured cells in CM media to induce the differentiation. The CM media contains 10% of FBS (fetal bovine serum), 1% of L-glutamine (cambrex), 1% of penicillin-streptomycin, 0.5μM of hydrocortisone, 5 μg/ml of insulin and 10 mM of nicotinamide. |
