Summary: | Vectors carrying PAS(plasmid achromobacter secretion) factor mutants are provided to improve expression of fused peptide or protein, and allow expressions of transmembrane proteins such as ryanodine receptor and human-originated protein such as calcipressin, that have been known not to be expressed in conventional vector systems. A PAS factor mutant comprises the amino acid sequence of SEQ ID NO:2. A nucleic acid molecule encoding the PAS factor mutant comprises the nucleotide sequence of SEQ ID NO:1. A vector contains the PAS factor mutant nucleic acid molecule. A method for mass producing a recombinant peptide or protein comprises the steps of: (a) transforming a host cell with a vector containing the nucleotide sequence encoding a recombinant peptide or protein fused with the PAS factor mutant nucleic acid molecule; (b) culturing the transformed host cell to express the recombinant peptide or protein; and (c) collecting the expressed peptide or protein, wherein the vector comprises the expression construct consisting of PAS factor mutant-encoding nucleotide sequence-fusion partner protein-encoding nucleotide sequence-recombinant peptide or protein-encoding nucleotide sequence. |
