Summary:A sustained-release pellet composition comprising venlafaxine hydrochloride is provided to improve uniformity of drug release rate and drug release control by using triple release control layers. A method for preparing the composition is also provided to improve convenience, reproducibility, rapidness and economic efficiency for preparation by carrying out the method in one device. A sustained-release pellet composition comprises: (a) 1.4-1.7 parts by weight of a core; (b) a first release control layer containing 1 part by weight of venlafaxine hydrochloride, and 0.02-0.9 parts by weight of a first release controller selected from cetostearyl alcohol, polyvinylacetate aqueous dispersion, dimethylaminoethyl methacrylate and neutral methacrylic acid copolymer, hydroxypropylmethylcellulose and methylcellulose; (c) a second release control layer containing 0.2-4 parts by weight of a second release controller selected from ethylcellulose and ethylcellulose aqueous dispersion; and (d) a third release control layer containing 0.25-1.3 parts by weight of a third release controller selected from ethylcellulose aqueous dispersion and polyvinylacetate aqueous dispersion, and is formulated as capsule.
