Summary: | PURPOSE: An electroporation-mediated DNA delivery method and a method for expressing erythropoietin using the same are provided, thereby improving the expression efficiency of a foreign gene in vivo and in vitro, and consequently improving the efficiency of gene therapy for various diseases. CONSTITUTION: An electroporation-mediated DNA delivery method comprises injecting a mixture of a transferring composition containing 50 to 150 mM of salts and DNA into a tissue, and subjecting the tissue to electronic stimulation, wherein the transferring composition is sodium chloride solution; the electronic stimulation is carried out by stimulating the tissue using electricity of 100 to 150 V/cm for 20 to 80 msec in 4 to 12 times; the tissue is striated muscle; the DNA is an animal cell expressing vector; the animal cell expressing vector is pJDK(KCCM 10343) comprising a foreign gene; and the foreign gene is erythropoietin gene.
본 발명은 전기적 자극을 이용한 DNA 전달방법 및 이를 이용한 에리스로포이에틴 발현방법에 관한 것으로, 보다 상세하게는 50 내지 150 mM의 염을 함유하는 전달 조성물 및 DNA를 포함하는 혼합물을 세포에 투여하고, 세포에 전기적 자극을 가하는 것을 포함하는 세포내 DNA 전달방법, 유전자 치료용 조성물 및 pJDK-mEPO 벡터를 제공한다. 본 발명의 DNA 전달방법은 외래유전자의 생체내, 외 발현효율을 증가시켜 각종 질병에 대한 유전자 치료 효율을 향상시킨다. |
