Summary:PURPOSE: A method for purification of human erythropoietin(EPO) is provided which rapidly, simply, and economically purifies human EPO from fluid using immunoaffinity chromatography. CONSTITUTION: Human EPO is purified from fluid using the first ion exchange chromatography, immunoaffinity chromatography, then the second ion exchange chromatography. In the first ion exchange chromatography, human EPO is bound at pH 8.0, and is eluted at pH 7.0-8.0 after other proteins are eluted at pH 4.0-4.5. In immunoaffinity chromatography, the fraction of human EPO is bound to human EPO-specific antibody bound column at pH 8.0, and is eluted at pH 2.0-2.5 after non-specific proteins are eluted at pH 7.0. In the second ion exchange chromatography, the fraction of human EPO is bound at pH 8.0, and is eluted at pH 7.0-8.0 after other proteins are eluted at pH 4.0-4.5. DEAE resin is used for the first and the second ion exchange chromatography.
