A positioning system and method of a mold exchanging machine are provided to improve work efficiency and productivity by securing a mold at the exact exchange position through full-automatic control. A positioning system of a mold exchanging machine includes a positioning device(30) adjusting a mold exchanger stopped at the exchange position to the exact position by the rotation-contact with a fixing device(20) and fixing it. The positioning device comprises a roller stopper(33) making rotation-contact with the fixing device, a hydraulic cylinder(35) including a cylinder loader(34) ascending/descending the roller stopper, a guide shift(36) preventing the bending of the cylinder loader when descending the roller stopper, and a guide plate(37) connecting the cylinder loader and the guide shift.
