A diagnostic formulation of Tsutsugamushi disease is provided to improve the convenience and sensitivity of detection by removing a cultivation process and detecting an antibody against Orientia tsutsugamushi in a biological sample. A diagnostic kit for Tsutsugamushi disease comprises (a) a sample pad for absorbing a biological sample, (b) a gold conjugation pad for binding to a human antibody in the sample, (c) a test membrane having a test line containing a mixed Tsutsugamushi antigen and a control line containing a control protein, and (d) an absorption pad for absorbing a remaining amount of sample, wherein the test membrane further has a pretest line containing the extract of Escherichia coli expressing no Tsutsugamushi antigen protein prior to the test line. 본 발명은 쯔쯔가무시 혼합 항원을 이용한 쯔쯔가무시병의 진단 키트에 관한 것이다. 구체적으로 상기 진단 키트는 시료가 흡수되는 샘플패드(sample pad), 시료 내의 인간 항체와 결합하는 금 결합 패드(gold conjugation pad), 쯔쯔가무시 혼합 항원을 포함하는 반응선(test line)과 대조군 단백질을 포함하는 대조선(control line)을 포함하는 반응막(test membrane), 및 잔량의 시료를 흡수하는 흡수패드(absorption pad)를 포함하는 테스트 스트립을 포함한다. 본 발명의 진단 키트를 이용하면 쯔쯔가무시 항체를 간단하게 검출할 수 있어 특별한 기계 조작 없이도 쯔쯔가무시 감염 여부를 육안으로 확인할 수 있다.
