Summary:PURPOSE:To obtain a DNA chain containing a biosynthetic gene for a carbapenem based antibiotic substance, by digesting the whole chromosomic DNA of Streptomyces.fulvoviridis with a restriction enzyme PstI. CONSTITUTION:The aimed DNA fragment of about 6.0kb mol. wt. obtained by digesting chromosomic DNA of Streptomyces.fulvoviridis N1501 with PstI, subjecting the resultant DNA fragment with a plasmid PIJ385 similarly treated with PstI to ligation treatment using a T4 ligase, introducing the ligation product into an FN24 strain, selecting a plasmid pCPG31 incorporating a biosynthetic gene of a carbapenem based antibiotic substance in a selection culture medium and then treating the above-mentioned plasmid with the PstI and having one recognition site with BglII, KpnI and SacI and tow recognition sites with BamHI and XhoI. Furthermore, a DNA fragment of about 1.7kb prepared by digesting the afore-mentioned DNA fragment with BamHI is also capable of imparting the ability to produce an antibiotic substance to a microorganism, such as FN24 or FN27, incapable of producing the antibiotic substance.
