Summary:PURPOSE:To accurately quantify an antigen, by forming an antigen-antibody composite by reacting an antigen to be measured in a specimen with the first antibody corresponding to said antigen and reacting the same with the second antibody corresponding to the antigen and irradiating the antigen-antibody composite with light to measure optical change quantity. CONSTITUTION:At first, the first antibody is allowed to act on a specimen containing an antigen to be measured to form an antigen-antibody composite. herein, as the first antibody, a monoclonal antibody or a polyclonal antibody corresponding to the antigen to be measured may be used. The first antibody is usually mixed with the specimen in a solution form and the concn. of the first antibody solution is about 5-30mg/ml. Next, the second antibody is added to the mixture of the specimen and the first antibody. As the second antibody, any one corresponding to the antigen to be measured may be used. From the point of time immediately after the second antibody is added, the reaction system is irradiated with light and the optical change quantity thereof is measured. When a calibration curve is formed on the basis of the specimens having known concns., the antigen in a specimen of unknown concn. can be quantified.
