Summary: | PURPOSE:To prepare the titled compound important as a synthetic intermediate of vitamin B1, in extremely high selectivity, by using a novel 4-alkoxy-1,3- dioxane-5-carbonitrile as a starting material, and reacting with acetamidine. CONSTITUTION:5-Acetamidomethyl-4-amino-2-methylpyrimidine is prepared by reacting a novel 4-alkoxy-1,3-dioxane-5-carbonitrile of formula (R is lower alkyl) (e.g. 4-methoxy-1,3-dioxane-5-carbonitrile) with acetamidine. Since acetamidine is an unstable substance, it is preferable to use in the form of a salt with an inorganic acid. The 4-alkoxy-1,3-dioxane-5-carbonitrile which is a novel compound used as a starting material can be prepared by reacting 3-alkoxy-2- propenenitrile compound with formaldehyde in the presence of an acid catalyst. |
