Summary:PROBLEM TO BE SOLVED: To provide polygalacturonase having glucose resistance, and to provide a method for producing the same. SOLUTION: This polygalacturonase is produced with a microorganism belonging to the genus Cryptococcus, and has the following physicochemical properties (1)-(9): (1) hydrolyzes the α-1,4,-bond of poly-D-galacturonic acid; (2) has the optimal pH of 3.5; (3) exhibits a stable enzymatic activity at a pH of 3.0-6.0, when held at 20 deg.C for 6 hr; (4) has the optimal temperature of about 22 deg.C; (5) exhibits a stable enzymatic activity of up to 25 deg.Cm, when held at a pH of 4.2 for 3 hr, and is gradually deactivated above the pH; (6) is not inhibited by mercuric chloride but scarcely by ethylenediaminetetra-acelic acid; (7) has a mol.wt. of about 70,000 dalton measured by a SDS-polyacrylamide gel electrophoresis method, (8) has an isoelectric point at a pH of 14.8, and (9) exhibits a relative activity of about 80% in a 500 mM glucose solution.
