PRODUCTION OF QUINOLINECARBOXYLIC ACID ESTER DERIVATIVE

PROBLEM TO BE SOLVED: To easily and safely produce a 1-cycloalkyl-7-fluoro-1,4-dihydro-4-oxo-3- quinolinecarboxylic acid ester derivative from an easily available raw material in a high yield and at a low cost by closing the ring of a 2-chloro-4-fluoro compound of the raw material. SOLUTION: An acry...

Summary:PROBLEM TO BE SOLVED: To easily and safely produce a 1-cycloalkyl-7-fluoro-1,4-dihydro-4-oxo-3- quinolinecarboxylic acid ester derivative from an easily available raw material in a high yield and at a low cost by closing the ring of a 2-chloro-4-fluoro compound of the raw material. SOLUTION: An acrylate ester derivative of formula I [R is a 1-7C alkyl; R is a (halogenated) cycloalkyl; X is H, F, Cl, a 1-7C alkyl of a 1-7C (halogenated) alkoxy; X is H, F or Cl; X is H, a 1-7C alkyl or nitro] [for example, 3-cyclopropylamino-2-(2-chloro-4,5-difluorobenzyl)acrylic acid ethyl ester] is subjected to a ring-closing reaction in the presence of a metal carbonate salt of the formula: Mn CO3 [M is an alkali metal or an alkaline earth metal; (n) is 1 or 2] in an aprotic polar solvent (for example, N,N- dimethylformamide) to produce the objective compound of formula II.
