Summary:PROBLEM TO BE SOLVED: To prepare a closed vesicle, used for a medicine such as an antitumor agent or an agent, etc., for diagnosis and having high safety by rehydrating a closed vesicle comprising a dehydrated micellar particle or a dehydrated amphiphilic micellar double layer under specific conditions. SOLUTION: This closed vesicle is prepared by rehydrating a closed vesicle comprising a dehydrated micellar particle or a dehydrated amphiphilic micellar double layer under low-temperature conditions, especially 10-0 deg.C when rehydrating the closed vesicle with a rehydrating solution. The closed vesicle can be applied to the one in which a medicine or an agent for diagnosis is introduced. The closed vesicle can especially be applied to an antitumor agent such as adriamycin, daunomycin or vinblastine and a pharmaceutically permissible salt and a derivative thereof as a medicine. A liposome can be cited as the closed vesicle. Furthermore, the liposome to which an antibody or a polyethylene glycol or both are applied is preferred.
