Summary:PROBLEM TO BE SOLVED: To obtain a new modified protein, comprising an amino acid sequence in which cysteine residue of a protein is converted into a lipid-nonbinding amino acid residue and useful as an antigen and a measuring reagent, etc., for an anti-Treponema pallidum antibody and for diagnosis, etc., of syphilis. SOLUTION: This new modified protein has a specific amino acid sequence in which cysteine residue of a protein is converted into a lipid-nonbinding amino acid residue such as alanine residue, glutamic acid residue or serine residue, etc., and is at least one of antigenic proteins having about 47kDa and about 17kDa molecular weight of a Treponema pallidum spirochete, etc., and useful as a measuring reagent for an anti-Treponema pallidum antibody, a diagnostic agent for syphilis and an antigen, etc., for producing the anti-Toriponema paridum antibody. The modified protein is obtained by extracting a DNA from the Treponema pallidum spirochete, amplifying the extracted DNA with a primer in which the cysteine residue of a gene of antigen having 47kDa molecular weight is substituted by alanine residue and expressing the resultant modified protein gene in a host cell.
