Summary: | PURPOSE: To obtain the subject new enzyme having lower ability to decompose cellobiose than that to cellooligosaccharides, capable of producting cellobiose and glucose from cellooligosaccharides, by improving its heat resistance through recombination of a part of an enzyme gene derived from Cellvibrio microbes. CONSTITUTION: This new heat-resistent β-glucosidase has an amino acid sequence containing an amino acid sequance of the formula, having the following characteristics: acting on cellulose degradation products to form glucose and cellobiose; having lower ability to decompose cellobiose then that to other cellooligosaccharides such as cellotriose, cellotetraose and cellopentaose; optimal pH: 6.0, stable at pH4-9; optimal temperature: 40-45 deg.C, stable up to 35 deg.C, and retaining 60% activity at 45 deg.C. This enzyme is obtained by recombination of a part of a β-glucosidase gene derived from Cellvibrio gilvus with a part of a β-glucosidase gene derived from Agrobacterium tumefaciens to impart a higher heat resistance followed by binding the resultant gene to a vector and then manifesting it in a host. |
