Summary: | PURPOSE:To separate a bluish green dyestuff in high efficiency by selectively isolating a great number of protoplasts prepared by treating callus of lavender with a specific cell wall dissolving enzyme and grinding. CONSTITUTION:Callus of lavender multiplied in MS medium containing a plant hormone such as kinetin is treated with a combined cell wall dissolving enzyme of 0.1-1.5wt.%, especially preferably 1% cellulase and 0.1-0.8%, especially preferably 0.2% pectolyase to give a great number of protoplasts, cells from which cell wall covering the outside of plant wall are removed in a viable state, which are selectively isolated. Since protoplasts are broken in a low isotonic solution such as ordinary medium or distilled water, a bluish green dyestuff contained in the cells is separated to break protoplasts. Consequently, the dyestuff is efficiently extracted. |
