Summary:To provide a screening method for a gene involved in blemish formation and a screening method for a substance that inhibits the formation of blemishes or a substance that ameliorates blemishes.SOLUTION: A method comprises: a step of preparing epidermal cells of an isolated pigment-enhanced site; a step of isolating DNA and RNA from the epidermal cells; a step of analyzing the DNA by whole-genome bisulfite sequencing; a step of analyzing the RNA by RNA-seq; and a step of selecting a gene using, as an index, an increase or decrease in DNA methylation compared to a prescribed standard and an increase or decrease of RNA expression compared to a prescribed standard.
