To provide a nucleic acid extraction method capable of reliably separating nucleic acids and impurities.SOLUTION: Provided is a nucleic acid extraction method. The method comprises: a first washing step for performing at least one time a first washing process in which an alcoholic solution is injected into a container equipped with a filter containing a specimen from which nucleic acids can be extracted and at the same time the container is pressurized to remove impurities other than the nucleic acids adsorbed on the filter to discharge the alcoholic solution from the container; a second washing-side pretreatment step in which a high-concentration alcoholic solution having a higher concentration than the alcoholic solution injected in the first washing step performed last in the first washing step is injected into the container, followed by a replacement process of replacing the inside of the container with the high-concentration alcoholic solution and discharging the high-concentration alcoholic solution; a second washing-side post-treatment step for performing a drying treatment for drying the high-concentration alcoholic solution after the second washing-side pretreatment step; and a nucleic acid recovery step of recovering the nucleic acid attached to the filter.SELECTED DRAWING: Figure 9 【課題】核酸と不純物を確実に分離できる核酸抽出方法を提供する。【解決手段】本発明の核酸抽出方法は、核酸を抽出可能な状態にされた検体を収容するフィルタ付きの容器にアルコール溶液を注入すると共に、前記容器を加圧して、前記フィルタに吸着する核酸以外の不純物を除去し、前記アルコール溶液を前記容器から排出する第一洗浄処理を少なくとも１回行う第一洗浄工程と、前記第一洗浄工程において最後に行われた前記第一洗浄処理で注入された前記アルコール溶液よりも高濃度の高濃度アルコール溶液を前記容器に注入し、前記容器内を該高濃度アルコール溶液に置換して、該高濃度アルコール溶液を排出する置換処理を行う第二洗浄側前処理工程と、前記第二洗浄側前処理工程の後に前記高濃度アルコール溶液を乾燥させる乾燥処理を行う第二洗浄側後処理工程と、前記フィルタに付着した前記核酸を回収する核酸回収工程と、を備える。【選択図】 図９
