Summary:To provide nucleic acid amplification apparatuses capable of performing real-time PCR with high-speed and having a small size portable on site, to provide plates for the apparatuses, and to provide nucleic acid amplification methods.SOLUTION: Provided is a reciprocal flow type nucleic acid amplification apparatus comprising: a heater that can form a denaturing temperature zone and an elongation/annealing temperature zone; a fluorescence detector capable of detecting transfer of the sample solution between the two temperature zones; a pair of liquid feeding mechanisms that enable transfer of the sample solution between the two temperature zones and that release the atmospheric pressure when the liquid feeding is stopped; a substrate on which a nucleic acid amplification chip can be placed; and a control mechanism that controls the drive of each liquid feeding mechanism by receiving an electrical signal from the fluorescence detector related to the transfer of the sample solution, the apparatus being characterized by performing real-time PCR by measuring the fluorescence intensity for each thermal cycle.SELECTED DRAWING: None 【課題】現場に持ち運び可能な小型で高速にリアルタイムPCRが可能な核酸増幅装置、該装置用のプレート及び核酸増幅方法の提供。【解決手段】変性温度帯と伸長・アニーリング温度帯を形成できるヒーター、前記2つの温度帯間の試料溶液の移動を検出可能な蛍光検出器、前記2つの温度帯間の試料溶液の移動を可能にし、かつ、送液停止時には大気圧開放される1対の送液用機構、核酸増幅用チップを載置可能な基板、試料溶液の移動に関する蛍光検出器からの電気信号が送られて各送液用機構の駆動を制御する制御機構を備え、サーマルサイクル毎の蛍光強度の計測を行うことでリアルタイムPCRを行うことを特徴とするレシプロカルフロー型の核酸増幅装置。【選択図】なし
