Summary: | To provide a methylation method of an aromatic compound, easily synthesizing a reaction substrate which becomes a raw material, and capable of being used as a manufacturing a [C] label tracer for PET.SOLUTION: By adding [C] labeled methyl iodide to an aprotic polar solvent in which a 0 valent palladium complex and a phosphine ligand are dissolved, as a first palladium complex formation process, [C] methyl palladium complex is formed. Further as a second palladium complex formation process, by adding aromatic halogen, where the halogen is I or Br, or aromatic triflate to an aprotic polar solvent in which the 0 valent palladium complex, the phosphine ligand, or an N-heterocyclic carbene ligand, an aromatic palladium complex is formed. By mixing a solution of the [C] palladium complex and a solution of the aromatic palladium complex, a [C] methyl group is bound to an aromatic substituent.SELECTED DRAWING: Figure 1
【課題】原料となる反応基質の合成が容易であり、PET用の[11C]標識トレーサーの製造方法として利用が可能な、芳香族化合物のメチル化方法を提供する。【解決手段】第1のパラジウム錯体形成工程として、0価のパラジウム錯体と、ホスフィン配位子とが溶解した非プロトン性極性溶媒中に[11C]標識ヨウ化メチルを加えて[11C]メチルパラジウム錯体を形成させる。さらに、第2のパラジウム錯体形成工程として、0価のパラジウム錯体と、ホスフィン配位子又はN-ヘテロサイクリックカルベン配位子とが溶解した非プロトン性極性溶媒に芳香族ハロゲン(ただし、ハロゲンはI又はBr)又は芳香族トリフレートを加えて芳香族パラジウム錯体を形成させる。そして、[11C]メチルパラジウム錯体の溶液と芳香族パラジウム錯体の溶液とを混合して芳香族置換基に[11C]メチル基を結合させる。【選択図】図1 |
