Summary: | To provide a manufacturing method of a solution of β-1,3-glucan capable of dissolving β-1,3-glucan, which is insoluble in neutral water at a triple helix state, in the neutral water without deteriorating physiological activity, biocompatibility or the like, β-1,3-glucan complex obtained by the method, and a pharmaceutical composition containing the same.SOLUTION: There is provided a manufacturing method of a solution of β-1,3-glucan in which 3 β-1,3-glucans form a complex having triplet helix structure in natural water, including: Process (1): a process for dissolving β-1,3-glucan, which has poor solubility in neutral water, in an alkali solution; Process (2): a process for mixing the alkali solution of β-1,3-glucan, and a buffer containing oligonucleotide or a derivative thereof of 1 wt.% to 80 wt.% of the β-1,3-glucan; and Process (3): a process for neutralizing the alkali solution containing the β-1,3-glucan, in order of (1), (2) and (3) or (1), (3) and (2).SELECTED DRAWING: None
【課題】三重らせん状態で中性の水に不溶なβ−1,3−グルカンを、生理活性や生体適合性等を損なうことなく中性の水に溶解させることを可能にするβ−1,3−グルカンの水溶液の製造方法、該方法を用いて得られるβ−1,3−グルカン複合体及びそれを含む医薬組成物を提供する。【解決手段】工程(1):中性で水に難溶性のβ−1,3−グルカンをアルカリ水溶液に溶解する工程、(2):前記β−1,3−グルカンのアルカリ水溶液と、当該β−1,3−グルカンの1重量%以上80重量%以下のオリゴヌクレオチド又はその誘導体を含む緩衝液とを混合する工程及び(3):前記β−1,3−グルカンを含むアルカリ水溶液を中和する工程を、(1)、(2)、(3)又は(1)、(3)、(2)の順序で含む、中性の水中で3本のβ−1,3−グルカンの分子鎖が三重らせん構造を有する複合体を形成しているβ−1,3−グルカンの水溶液の製造方法。【選択図】なし |
