Summary: | PROBLEM TO BE SOLVED: To provide methods for selecting cells with zero fucose level useful as a host cells for expressing recombinant proteins.SOLUTION: Provided is a method comprising the steps of: introducing genetic mutations into a population of Chinese hamster ovary (CHO) cells by contacting the cells with methotrexate (MTX); contacting the population of CHO cells including mutated cells with a non-toxic fucose binding agent for a period of time that allows the binding agent to bind to the fucose moiety on the cell membranes of the cell population but not to kill the cells; and eliminating a subpopulation of cells bound to the fucose binding agent thereby selecting cells having zero fucose content useful as host cells for recombinant protein expression. The invention also provides cells and cell lines useful as host cells for expressing recombinant proteins.SELECTED DRAWING: Figure 1
【課題】組換えタンパク質を発現するための宿主細胞として有用な、ゼロフコースレベルを有する細胞を選択する方法の提供。【解決手段】方法は、メトトレキサート(MTX)に細胞を接触させることにより遺伝的突然変異をチャイニーズハムスター卵巣(CHO)細胞の集団に導入する工程と、突然変異細胞を含むCHO細胞の集団を、非毒性フコース結合剤と、該フコース結合剤が、細胞集団の細胞膜上のフコース部分に結合するのに充分な時間接触させる工程と、その時間が細胞の殺傷を可能としない時間である工程と、突然変異細胞を含む細胞の集団から、フコース結合剤に結合する細胞の亜集団を除去し、それにより組換えタンパク質の発現のために宿主細胞として有用な細胞を選択する工程と、選択された細胞はゼロフコース含量を有する工程と、を含む製造方法。また、組換えタンパク質を発現するための宿主細胞として有用な細胞および細胞株。【選択図】図1 |
