Summary:PROBLEM TO BE SOLVED: To provide composite proteins including antibodies which show reduced immunogenicity, in particular, composite antibodies for use in humans, antibodies modified by removing one or more T cell epitopes and generation methods thereof.SOLUTION: A composite antibody variable region or an antigen binding fragment thereof is characterized by: (1) the variable region comprises a heavy chain sequence and a light chain sequence, each of which comprises plural segments of an amino acid sequence of 2 to 31 amino acids long derived from one or more other antibodies or antigen binding fragments thereof, where the heavy chain segments and the light chain segments are selected from specific sequences, the segments are neither whole CDRs nor whole framework regions; (2) the combined antibody variable region or the antigen binding fragment binds to human TNFα; and (3) the final composite antibody variable region contains no T cell epitope.SELECTED DRAWING: None 【課題】低免疫原性を示す抗体を含む合成タンパク質の提供。特に、ヒト用複合抗体を提供し、一又はそれ以上のT細胞エピトープを除去するように修飾された抗体、及び、その生成方法の提供。【解決手段】合成抗体可変領域又は合成抗体可変領域の抗原結合フラグメントであって、(1)前記可変領域が重鎖配列及び軽鎖配列を含み、前記重鎖配列及び軽鎖配列の各々が1または複数の他の抗体又は抗体の抗原結合フラグメント由来の2乃至31アミノ酸長のアミノ酸配列の複数のセグメントを含み、前記重鎖セグメント及び軽鎖セグメントが特定の配列から選択され、前記セグメントが全CDRでも全フレームワーク領域でもなく;(2)前記合成抗体可変領域又は前記抗原結合フラグメントがヒトTNFαに結合し;及び(3)最終合成抗体可変領域にT細胞エピトープを含まない;ことを特徴とする合成抗体可変領域又は合成抗体可変領域の抗原結合フラグメント。【選択図】なし
