Summary:PROBLEM TO BE SOLVED: To provide a 2'-deoxynucleoside (dNS) production method suitable for industrial production.SOLUTION: The invention relates to a method for producing dNS via 2-deoxyribose (dR) by performing at least one of the following (i), (ii), and (iii): (i) subjecting divalent metal salt of 2-dehydro-3-deoxy gluconic acid (KDG) to decarboxylation response to synthesize dR, and synthesizing dNS using the obtained dR as a reaction intermediate; (ii) synthesizing dR from KDG or a salt thereof using a variant enzyme having dR synthesis ability, and synthesizing dNS using the obtained dR as a reaction intermediate; or (iii) synthesizing 2-deoxyribose-5-phosphate ester (dR5P) from dR using the variant enzyme having dR5P synthesis ability, and synthesizing dNS from the obtained dR5P.SELECTED DRAWING: None 【課題】工業生産に適した2'−デオキシヌクレオシド(dNS)の製造方法を提供する。【解決手段】本発明は、少なくとも下記(i)、(ii)及び(iii)のいずれか1つを実行することにより、2−デオキシリボース(dR)を経由してdNSを製造する方法である。(i)2−デヒドロ−3−デオキシグルコン酸(KDG)の二価金属塩を脱カルボキシル化反応に付してdRを合成し、得られるdRを反応中間体としてdNSを合成する;(ii)dR合成能を有する変異型酵素を使用して、KDG又はその塩からdRを合成し、得られるdRを反応中間体としてdNSを合成する;(iii)2−デオキシリボ−ス−5−リン酸エステル(dR5P)合成能を有する変異型酵素を使用してdRからdR5Pを合成し、得られるdR5PからdNSを合成する。【選択図】なし
