PROBLEM TO BE SOLVED: To provide macrocyclic compounds, and to provide methods of making and using the same.SOLUTION: This invention relates to compounds useful as antiproliferative agents and/or anti-infective agents, wherein these compounds are useful as, for example, antineoplastic agents, antimicrobial agents, antibacterial agents, antfungal agents, antiparasitic agents, and antiviral agents, without limitation. This invention also relates to compounds useful as anti-inflammatory agents and/or prokinetic agents, without limitation. For example, the anti-inflammatory agents are used in treating chronic inflammatory airway diseases; and the prokinetic agents are used in treating gastrointestinal motility disorders such as gastroesophageal reflux disease, gastroparesis (diabetic and post surgical), irritable bowel syndrome, and constipation. Further, the compounds can be used to treat or prevent a disease state in a mammal caused or mediated by a nonsense or missense mutation.
