Summary:PROBLEM TO BE SOLVED: To provide an industrially practicable method having excellent operability for producing optically active L-or D-tert-leucine important as a raw material for medicines and agrochemicals. SOLUTION: In a method for making a biocatalyst for stereoselectively hydrolyzing tert-leucine amide act on tert-leucine amide and then substituting a solvent of a reaction solution of the biocatalyst from a water system to a poor solvent system of optically active tert-leucine so as to crystallize, precipitate and collect formed optically active tert-leucine, the solution to be supplied to the solvent substitution contains an acetate radical and an alcohol is used as a poor solvent when substituted with a poor solvent system.
