Summary:PROBLEM TO BE SOLVED: To provide a method for amplifying a nucleic acid capable of suppressing a burst of a sample such as a reaction liquid in a nucleic acid amplification reaction such as a PCR, to provide a device for amplifying the nucleic acid suitable for use in the above method, and to provide a method for holding a minute amount of a liquid suitable to the application in the above amplification method. SOLUTION: The method for amplifying a nucleic acid includes: arranging a liquid-holding means where a reaction liquid to conduct a nucleic acid amplification is held without being sealed in an atmosphere under pressure; and performing the nucleic acid amplification reaction with the boiling point of the reaction liquid at 105°C or higher. The nucleic acid amplification device for use in such a nucleic acid amplification method is provided, including: a means for adjusting the temperature of the reaction liquid; a sealed chamber installed with the liquid-holding means holding the reaction liquid without sealing the reaction liquid; and a pressure means connected with the sealed chamber to pressurize the inside of the sealed chamber. The method for holding a minute amount of a liquid is also provided, including steps of: holding the minute amount of the liquid on a substrate: covering the minute amount of the liquid with a covering liquid that is smaller in specific gravity than the minute amount of the liquid, is not admixed with the minute amount of the liquid, and suppresses the evaporation of minute amount of the liquid; installing the substrate in the sealed chamber; and heating the minute amount of the liquid in such a condition that the inside of the sealed chamber is pressurized.
