Summary: | PROBLEM TO BE SOLVED: To provide a pretreatment method for detecting an acid-fast bacterial gene contained in expectorated sputum, which is quick and accurate, has high yield, will not cause scattering or contamination of liquid droplets in a closed system, and which isolates acid-fast bacteria from expectorated sputum, in a ready and safe manner, and to provide a method for detecting an acid-fast bacterial gene which includes treating acid-fast bacteria by the pretreatment method, extracting a gene that specifically amplifies and detects the gene. SOLUTION: The pretreatment method for detecting an acid-fast bacterial gene, contained in expectorated sputum includes adding a solubilizing agent of expectorated sputum and a treatment agent for removal of various germ to expectorated sputum, and directly binding/collecting acid-fast bacteria by a solid substrate or a nonspecific ligand which is immobilized on the solid substrate without having to remove the treatment agent.
