Summary: | PROBLEM TO BE SOLVED: To provide a common method applicable to all variants generally used as a DNA marker for genetic analysis. SOLUTION: The analysis method comprises preparing a polynucleotide target involving nucleotide of variable base and one oligonucleotide probe fixed on a support which probe is complementary to the target and terminates at a position before its 5' terminal by one base; and comprises (a) incubating the target with the probe to form a duplex chain, (b) incubating the duplex chain with a labelled oligonucleotide complementary to the target including predicted variant of the target under a ligation condition, and (c) monitoring the addition of oligonucleotide to probe by ligation of the step (b) as one exhibiting point mutation at the specific position in the target.
