Summary:PROBLEM TO BE SOLVED: To provide a method for observing tissue structure, capable of accurately adapting to the transparency of objects to be measured and observing tissue structures. SOLUTION: In the method for observing tissue structure for observing the tissue structure of an object to be measured 6 through the use of an optical microscope device 20, the distance between a condenser lens 2 and a reference point 15 in the optical axis is determined. On the basis of the relation expressed by the expression y=tanθ×(x-x0)(where y is the distance between an pattern edge 9d of a spatial diaphragm 9, installed on a diffraction image surface 8 and the optical axis 7) previously determined with respect to the distance x and the scattering angle θ of the scattered light, y is computed. The spatial diaphragm 9, having the pattern edge of y of which the distance from the optical axis 7 is computed, is installed on the diffraction image surface 8. Then the focus of the object lens 10 is made to coincide with the object to be measured 6 and the object to be measured 6 is observed. The intensity of the scattered light at scattering angles within a specific range passing through the spatial diaphragm 9 and the intensity of scattered light at full scattering angles with the spatial diaphragm 9 removed are measured.
