Abstract PROBLEM TO BE SOLVED: To provide a method and a device capable of resolving a problem of an over range without physically using, for example, two detectors, and capable of detecting an enzyme even when a substrate to be converted into a detectable substance by the enzyme becomes short due to an extremely high quantity of the enzyme to be determined. SOLUTION: In the method and device for determining the quantity of the enzyme, a reaction liquid is produced by adding the enzyme to a solution of the substrate converted into an optically measurable substance by enzyme activity of the enzyme, an inhibitor suppressing the enzyme activity of the enzyme is added to the reaction liquid, and one or more optical measurement is carried out in a state suppressing the enzyme activity of the enzyme.
