Summary:PROBLEM TO BE SOLVED: To obtain an administration composition capable of releasing most of a contained drug in a short time from the start of the drug release irrespective of the length of lag time, retained in the release site and maintaining a high local concentration as compared with that of a usual pharmaceutical preparation. SOLUTION: This pharmaceutical composition comprises a core composed of the drug and a swelling agent and a coating membrane layer around the core and is characterized in that (a) the coating membrane layer is composed of a water-insoluble and water-permeable substance and a part of the coating membrane layer has (1) a smaller membrane thickness than that of another coating layer part or (2) a lower breaking strength to drawing of the coating membrane layer accompanied by the swelling of the core and (b) the drug in the core is released to the outside of the composition by causing cracking in a part of the coating membrane layer by swelling of the swelling agent after the passage of a prescribed time from the oral administration to a warm- blooded animal.
