PROBLEM TO BE SOLVED: To provide a method for producing a useful substance using a Hordeum Vulgare D-hordien promoter in a seed of Hordeum Vulgare and a seed of the transformed Hordeum Vulgare obtained by the method. SOLUTION: The invention provides the method for producing the substance in the seed of Hordeum Voulgare containing the following processes and comprising using the Hordeum Vulgare D-hordien promoter (pDPP3) described in sequence number 1 of the sequence table in the specification: (1) a process transducing the Hordeum Vulgare D-hordien promoter and a gene cassette containing a gene of the promoter positioned in the downstream into a protoplast of the Hordeum Vulgare; (2) a process culturing the protoplast transduced with the gene cassette and obtaining a transformed cell; (3) a process making the transformed cell redifferentiate and obtaining a redifferentiated plant; and (4) a process making the redifferentiated plant ripen and obtaining the seed of Hordeum Vulgare.
