Summary:PROBLEM TO BE SOLVED: To obtain an oligonucleotide of high chiral purity bearing a phosphorothioate linkage. SOLUTION: This oligonucleotide is obtained by chemical and enzymatical methods for synthesizing a phosphorothioate oligonucleotide in which all the nucleotide units are substantially connected together by either all the Sp phosphorothioate saccharide linkages or all the Rp phosphorothioate saccharide linkages, preferably a phosphorothioate oligonucleotide complementary to at least a part of target RNA or DNA sequence and a sequence peculiar phosphorothioate oligonucleotide bearing a substantially chirally pure saccharide linkage.
