Summary:PROBLEM TO BE SOLVED: To provide a method for rapidly and accurately assaying the stability of the DNA transduced into an animal cell, and further to provide an animal cell vector suitable for the use in the method. SOLUTION: A vector having both of a gene resistant to a first medicine such as an antibiotic, and a gene sensitive to a second medicine such as cyclophosphamide is used. The vector can simultaneously produce a resistant gene product and a sensitive gene product from one mRNA transcribed from the same promoter, and, if necessary, can have Ori functioning in the animal cell. The animal cells having the transduced vector is cultured in a medium containing the first medicine to select the animal cells having the vector, and the selected cells is cultured in a medium containing no medicine. The cultured animal cells are cultured again in a medium containing the second medicine to select the animal cells having lost the vector. The numbers of colonies are counted to assay the stability of the DNA.
