Summary:Methods and reagents for identifying antigens in tissues on the basis of a cytochemical immunoenzyme method are disclosed. Methods and reagents are based on an antibody which is specific for the antigen to be identified, beta -D-galactosidase or beta -D-galactosidase conjugate and a chromogenic substrate specific for beta -D-galactosidase. Particularly with regard to reliability of colour and constancy of colour, the method of the present invention is superior to the peroxidase PAP method which is most commonly used at present in the area of immunoenzymatic cytochemical techniques.
