1476116 Triazole derivatives CIBA-GEIGY AG 9 June 1975 [14 June 1974] 24616/75 Heading C2C Novel compounds of Formula I wherein R 1 is H, C 1-7 alkyl, hydroxymethyl optionally etherified or esterified, formyl, carboxy optionally functionally modified, or aminomethyl optionally mono or disubstituted; B 2 is C 1-7 alkyl and R 3 is H or C 1-2 alkyl or NH 2 R 3 is a saturated monoheterocyclic ring and rings A and B may be substituted by halogen, CF 3 , NO 2 , C 1-7 alkyl or C 1-7 alkoxy and salts thereof are prepared by one of the following methods: (a) a reactive ester of a compound of Formula II is reacted with HNR 2 R 3 or an alkali metal derivative thereof; (b) a compound of Formula IV is reacted with formaldehyde or formic acid; (c) a compound of Formula V is oxidized; (d) a compound of Formula VI wherein R 6 is C 1-7 alkyl is treated with a base and optional conversion to a salt or a substituent group is converted to a derivative thereof by standard procedures. Reaction esters (mainly halides) of compounds of Formula II above are prepared by halogenation and optional conversion of functional groups to their derivatives. Intermediates of Formula V are prepared by NaBH 4 reduction of the ketones optionally followed by conversion of functional group to their derivatives. [2 - (Dimethylamino) - acetamido] - [2 - (ochlorobenzoyl) - 4 - chlorophenylazo] - malonic acid diethyl ester is prepared by reacting 2- amino - 21,5 - dichlorobenzophenone which has been diazotized with [2-(dimethylamino)acetamido]-malonic acid diethyl ester. Pharmaceutical compositions in conventional forms for oral, rectal or parenteral administration and having anticonvulsant activity comprise an above novel compound and a carrier or diluent.
