Summary:An isothermal process for amplifying a double-stranded nucleic acid target molecule that relies on upstream and downstream primers and an oligonucleotide, wherein at least a portion of the oligonucleotide is complementary to a portion of the target sequence intervening the upstream and downstream primers, wherein the upstream and downstream primers do not amplify the target molecule independently of the oligonucleotide and wherein the oligonucleotide facilitates the separation of the double-stranded nucleic acid target molecule thereby allowing the primers access to bind to complementary single-stranded DNA on the target molecule.
